A) Otilonium is the prototype of a class of 2-aminoethyl-N-benzoylamino-benzoate quaternary salts which possess antispasmodic properties in the gastrointestinal tract, particularly in the colon, at doses which do not affect gastric secretion or produce typical atropine-like adverse effects.

B)Otilonium acts predominantly by modifying Ca(2+) fluxes from cellular and extracellular sites together with a direct action on the contractile proteins, and (at high concentrations) on some enzyme activities such as (Ca(2+)- Mg(2+))-ATPase and (Na+-K+)-ATPase.
C) Binding studies carried out in the rat colon showed that otiloniume competitively interacts with the verapamil binding site on L-type Ca(2+) channels and muscarinic M2 receptors with an IC50 of 1020 and 1220 nM, respectively.
D)Under in vitro conditions, otilonium was found to block cholinergic nicotinic receptors in bovine chromaffin cells.
